Acute brain slices for electrophysiology must be cut fresh, without freezing or embedding. A blade oscillating at 50–100 Hz and 0.1–3 mm amplitude advances at 0.01–1.5 mm/s, keeping vertical deflection under 1 µm. Sections run 20–500 µm on specimens up to about 40 mm across, with ice jacket or Peltier bath cooling.
